Description: URB-597 is a potent and selective inhibitor of the enzyme fatty acid amide hydrolase (FAAH), which is involved in the hydrolysis of endogenous cannabinoids such as anandamide. Inhibition of FAAH may serve to enhance the analgesic effects of endogenous cannabinoids. URB-597 has been shown to inhibit FAAH in rat cortical neuron membranes and in intact neurons with IC50 values of 4.6nM and 0.5nM, respectively. The compound has also been shown to have anxiolytic effects in rats (ID50=0.15mg/kg). Because URB-597 acts on FAAH and does not interfere with the binding of anandamide to the CB1 and CB2 receptors, it is a novel tool for drug development.

Description: VASP (vasodilator stimulated phosphoprotein) is a proline-rich protein substrate of cAMP- and cGMP-dependent protein kinases. Phosphorylation of VASP at Ser-157 causes a mobility shift in SDS gel electrophoresis from 46 to 50 kDa, which has been used as a convenient marker to monitor cyclic nucleotide-dependent protein kinase activity. VASP is the founding member of the Ena-VASP protein family, comprising the Drosophila protein Enabled (Ena), its mouse homologue Mena (mammalian Enabled), and mouse EVL (Ena-VASP-like protein). With these proteins VASP shares a conserved overall domain organization:
a) the conserved N-terminal Ena-VASP homology domain 1 (EVH1), which mediates binding to a proline-rich motif,
b) a more divergent proline-rich central domain (which is responsible for profilin binding), and
c) a conserved C-terminal EVH2 domain.

Particularly high VASP levels are present in platelets, although VASP is expressed in a wide variety of cell types and tissues. In cultured cells, VASP is associated with focal adhesions, cell-cell contacts, microfilaments, and highly dynamic membrane regions. From in vitro binding data VASP has been suggested to link profilin to zyxin, vinculin, and the Listeria spp. surface protein ActA, respectively. Functional evidence indicates that VASP is a crucial factor involved in the enhancement of actin filament formation and the actin-dependent motility of intracellular bacterial pathogens.

Description: Mammalian apurinic/apyrimidinic (AP) endonuclease (APE1; APEX; HAP1; Ref-1) is a multifunctional, bipartite enzyme that belongs to the class II AP endonucleases and that plays an important role in numerous, cellular functions, like repairing abasic sites (loss of purine or pyrimidine base) in DNA, regulating the redox state of other proteins that play roles in oxidative signalling, transcription factor regulation (Fos, Jun, NF-κ B, Myb, HIF-1α, CREB, Pac), cell cycle control (p53) and apoptosis. APE1 initiates the DNA base excision repair (BER) by cleaving the DNA immediately adjacent to the 5’ of an AP site to produce a hydroxyl group at the 3’ terminus of an unmodified nucleotide upstream of the nick and a 5’-deoxyribose phosphate moiety downstream. The product of APE1 is further processed by DNA polymerase β to release 5’-deoxyribose phosphate with its intrinsic lyase activity. The nicked DNA is then sealed by DNA ligase I or DNA ligaseIII/XRCC1 to complete this repair process. In addition to the AP endonuclease activity, APE1 also possesses a 3’-5’ DNA exonuclease activity, a 3’-phosphodiesterase activity, a 3’-phosphatase activity, and a RNase H activity. It has been shown that APE1 is capable of excision L-configuration deoxyribonucleoside analogs from the 3’ termini of DNA. Human APE1 gene is located on chromosome 14q 11.2-12.
