CleanCap® Reagent AU is designed for the co-transcriptional capping of self-replicating or self-amplifying (saRNA) to produce an optimal Cap 1.
Designed specifically for co-transcriptional capping of saRNA using in vitro transcription

Produces saRNA with a 5′-AU cap found in the (+) strand alphavirus genomes and is designed to be compatible with the natural alphavirus sequence

CleanCap Reagent AU is specifically designed for saRNAs based on the genomes of positive sense (+) strand RNA viruses such as Venezuelan equine encephalitis virus (VEEV), Semliki forest virus (SFV) and Sindbis virus (SIN). The (+) strand genomes of these viruses start with a 5′-AU.
Cap-1 saRNAs produced by CleanCap AU are more suitable for eukaryotic usage than Cap-0 saRNAs produced by legacy co-transcriptional capping methods such as anti-reverse cap analog (ARCA). In addition, compared to enzymatic capping, capping with CleanCap AU analog offers more streamlined and efficient manufacturing of saRNAs.
CleanCap AU can be used in conjunction with TriLink’s modified and unmodified NTPs, as well as in vitro transcription (IVT) enzymes. Using the recommended conditions, transcription with CleanCap AU results in >95% capped material, a Cap-1 structure, and crude yields of 4 to 6 mg/ml of transcription.
CleanCap Reagent AU requires an AU initiator on DNA template.
