Digitonin is a mild nonionic detergent. It permeabilizes plasma membranes of eukaryotic cells by complexing with membrane cholesterol and other unconjugated β-hydroxysterols.

Digitonin has used for cholesterol determination, use of digitonin-permeabilised adipocytes for cAMP studies, solubilisation of membrane proteins. The water soluble form can be used for the isolation of mitotic chromosomes. Mild nonionic detergent used to solubilise receptors and permeabilise cellular and nuclear membranes.
Soluble in boiling water (1g/10 ml). Heat for 15 minutes at 98 °C. Cool and place at 4 °C for 3 hrs and filter off any precipitate. Soluble in absolute alcohol (1 g / 57 ml) and 95% alcohol (1 g).
