Human CD4+/CD25+/CD127- Regulatory T cells from peripheral blood

CD4+/CD25+ regulatory T cells were isolated from leukapheresis product by CD127 depletion and positive selection using CD25 immunomagnetic beads. Peripheral blood cells are collected from normal human volunteers by direct collection or by using the COBE® Spectra or Spectra Optia® Apheresis System where cell-rich leukapheresis product is extracted using continuous flow centrifugal technology directly into the collection bag.

  • Test negative for Hepatitis B and C, HHIV 1/2, HTLV-I/II, HIV-1/HCV/HBV Nucleic Acid, WNV Nucleic Acid, Trypanasoma cruzi and Syphilis
  • Cryopreserved in CryoStor® CS10 media

CD4+/CD25+/CD127- regulatory T cells (or dim regulatory T cells) express intracellular transcription factor FoxP3. They are critical to the maintenance of immune cell homeostasis and peripheral tolerance.The immunosuppressive mechanism of regulatory T cells is elusive, including the secretion of immunosuppressive soluble factors such as IL-9, IL-10 and TGF beta, cell contact mediated regulation via the high affinity TCR, and other costimulatory molecules such as CTLA-4, GITR, and cytolytic activity, among others. Understanding the mechanisms of T Reg Cells brings broad implications for the development of therapeutic strategies for many disease processes, including cancer, diabetes, and immune mediated diseases.

Storage: Either prepare cells for long term storage in liquid nitrogen vapour phase or thaw for use. Storage in liquid phase nitrogen is NOT recommended. Short term storage of cells (less than 2 weeks) at –80 °C is acceptable, but should be minimised to ensure maximum stability. Once thawed, samples must be used immediately.

Caution: Intended for only research use and not for direct therapeutic or diagnostic use in human or animals, or for further manufacturing of therapeutic products or devices. And storage in liquid phase nitrogen is not recommended.
