Gram-colour staining kit, containing 500 ml each of crystal violet solution, Lugol's solution stabilised, Safranin solution and 2×500 ml decolourisation solution.
In bacteriology, Gram staining allows a fast differentiation of bacteria in Gram-positive and Gram-negative. The murein structure of the bacteria wall is the basis of the colour affinity. In the first step, bacteria is stained with crystal violet, an aniline dye. After treatment with iodine solution (Lugol’s solution), a dye-iodine complex is formed. During the decolourising step, this complex stays in the multi-layer murein structure of the cell wall of Gram-positive bacteria - they appear blue. Gram-negative bacteria have a monolayer murein structure, the dye-iodine complex does not stay bound to the cell wall - they are decolourised. Gram-negative bacteria can then be counterstained with safranin solution and this will then appear orange. The Gram-colour stain set contains 5×500 ml safe PE bottles and is sufficient for staining up to 250 slides. It fulfils all quality requirements for IVD registration and CE certification.
